I did the ES Q and then the RX Q. I don't know if it mattered. I also had the old packaging of diapers but I had a mixture or the regular supreme and pull ups. Maybe the registers are programmed different or something.

I don't think it matters but they've always done ES then Rx for me. The Rx one has issues with size 1 & 2 packs because they have more than 40 ct (the coupon specifies less than 40 ct). If you need 1's & 2's then maybe buy 3's and then exchange them right then and there after buying them?
